Abstract / short description:
Abortions are defined as spontaneous pregnancy loss before 24 weeks of gestation. At least 25%, probably up to 50% of women suffer from abortion. During pregnancy, maternal immune cells are in close contact to cells of the semi-allogenic fetus. Maternal immune system has tightly to be regulated to avoid rejection of the fetus. Myeloid derived suppressor cells (MDSC) are myeloid cells with suppressive activity on other immune cells, especially on T-cells. Originally, MDSC were described under tumor conditions, where they inhibit anti-tumor immune responses. Our results show that MDSC accumulate also under physiologic conditions during pregnancy in maternal and fetal organism and that they play a role in maintenance of maternal-fetal tolerance. Until now, it is unclear, which factors regulate accumulation and function of MDSC during pregnancy. Human leucocyte antigen G (HLA-G) is an immuno-modulatory MHC class I molecule expressed at the maternal-fetal interface. Qa2 is the murine homologue of HLA-G. In this project, we will analyze the impact of HLA-G and Qa2 on accumulation and function of MDSC during pregnancy and on pregnancy maintenance with the aim to develop an immunological therapy for recurrent abortions.
