Abstract / short description:
Our aim is to structurally characterize the interaction of polySia with human Siglec-11 and murine Siglec-E to identify a recognition pattern of glycan attachment. In a similar approach, we will focus on the structural characterization of polySia binding to the chemokine CXCL12. For both targets, we will identify crucial glycan binding amino acids and determine their influence on the affinity by site-directed mutagenesis in combination with in vitro binding studies.
