Abstract / short description:
The central aim of this project is to confirm and validate that vaccination of AAD mice with biodegradable microspheres protects them from infection with influenza viruses (IV). Experiments from a previous collaboration between the research groups of Prof. Groettrup at the University of Konstanz and Prof. Planz at the University of Tübingen will be confirmed that two highly conserved synthetic peptides of IV together with an immunostimulant (riboxxime) can be encapsulated in microspheres made of the polymer poly(D,L-lactate-co-glycolate) (PLGA-MS) in such a way that all three pharmacologically active agents are released in a controlled manner in aqueous medium. These PLGA-MS will then be used to vaccinate AAD mice to confirm that strong antiviral immune responses consisting of cytotoxic T lymphocytes (CTL) are generated by this vaccination in the lungs and spleens of these mice. The aim is to validate that co-encapsulation of two IV peptides generates equally strong immune responses against both peptides as when these peptides are encapsulated separately in PLGA-MS. Furthermore, it will be investigated over a period of four months how long after vaccination the CTL response against the two peptides is maintained and whether booster vaccination enhances the CTL response again.
