Abstract / short description:
Regulation of proteins at the plasma membrane is the prerequisite for cellular development and viability. Regulation of these proteins occurs by their selective targeting for vacuolar degradation.
This mechanism thus allows for rapid responses to extracellular stimuli during plant-pathogen interactions or environmental changes. Degradative regulation of membrane proteins is initiated by ubiquitination, which triggers internalization and transport to the early endosome/trans-Golgi network. Further transport leads via endosomal maturation to multivesicular bodies that finally fuse with vacuoles.
Delivery of membrane proteins to the vacuolar lumen necessitates their relocation from the limiting endosomal membrane into intralumenal vesicles, resulting in the multivesiculate morphology of late endosomes. This relocation requires the function of three endosomal sorting complexes required for transport (ESCRT)-complexes and further ESCRT-associated proteins.
In this joint proposal, we aim at the characterization of the sequence of action of the ESCRT machinery that starts with the cargo-recognition and proceeds via further cargo-ESCRT- and ESCRT-ESCRT-interaction to the formation of intralumenal vesicles of endosomal compartments.
